Dramatic footage shows thick black smoke pouring out of the window of a third-floor flat in Turin Street, Bethnal Green.
London Fire Brigade said a man suffering from smoke inhalation has been taken to hospital.
Residents are believed to have been evacuated from the building.
Ten fire engines and 72 firefighters have been sent to the scene to battle the flames.
The flat and the building’s roof were destroyed by the flames.
The fire brigade received nearly 50 calls about the blaze.
The London Fire Brigade spokesperson said: ‘The cause of the fire is under investigation.’
Firefighters were called at 4.25pm and the blaze was under control by 7.30pm.
